MRM & SRM of Mitochondrial Proteomics

Selected reaction monitoring (SRM), also known as Multiple Reaction Monitoring (MRM) is a targeted quantitative proteomics method for the study of target protein molecules. Based on the information of the target molecule, MRM/SRM selects the data to collect the mass spectrum data, collects the signal that conforms to the rules of the target ion, and removes the interference of the ion signal that does not conform to the rule.

MRM/SRM mass spectrometry analysis goes through three stages. First, the parent ions which are consistent with the specificity of the target molecule are screened by MS. Secondly, these parent ions are broken by collision to remove the interference of other ions. Finally, only the selected specific MS/MS2 ions are collected by mass spectrometry. MRM/SRM mass spectrometry is a highly accurate technique for quantitative identification of proteins, and it is an excellent method for a one-time accurate quantitative study of multiple target proteins in complex samples. If the target peptide labeled by isotope is used as the internal reference, the absolute quantitative identification of protein can be achieved.

MRM/SRM monitors each precursor ion/product ion transition at a time, while PRM analyzes all productions derived from a precursor ion in concert with high resolution and mass accuracy. Both MRM/SRM and PRM allow relative and absolute quantification of proteins, peptides, and metabolites.
